Nectin, Membrane Protein Needed for Attachment of A TPase
نویسندگان
چکیده
We have isolated from Streptococcus faecalis plasma membranes a new protein, termed nectin, which is required for the attachment of the membrane enzyme, ATPase. Both nectin and membrane-bound ATPase are released from the native membranes by a procedure, described previously, which depends on repeated washing in dilute buffer without Mg++. Resolution of the solubilized nectin and ATPase can then be accomplished by gel filtration through agarose. The ATPase, separated from neck by passage through agarose, fails to reattach to enzyme-depleted membranes unless nectin is added. The amount of ATPase that recombines with the depleted membranes is proportional to the amount of nectin added until the membrane receptor sites for the enzyme become nearly saturated. Nectin is heat labile and has an ultraviolet absorption spectrum typical of a protein. By the method of gel filtration through Sephadex, we estimate that its molecular weight is 37,000.

Nectins belong to a family of immunoglobulin (Ig)-like cell-adhesion molecules comprising four members, nectin-1 through nectin-4. Nectins are involved in formation of the mechanical adhesive puncta adherentia junctions of synapses. Nectins share the same overall structural topology with an extracellular region containing three Ig modules, a transmembrane region, and a cytoplasmic region. In ne...
متن کاملDominant transposition-deficient mutants of maize Activator (Ac) transposase.
The maize transposable element Activator (Ac) encodes a transposase (TPase) protein, whose DNA-binding domain is located in a basic region around aa 200. The N-terminal 102 aa of the TPase are not required for the transposition reaction. In transfected petunia protoplasts, we analyzed the protein levels of the N-terminally truncated TPase and mutants thereof and the corresponding transposition ...
متن کاملNectin Couples Cell-Cell Adhesion and the Actin Scaffold at Heterotypic Testicular Junctions
Actin-based cell-cell adherens junctions (AJs) are crucial not only for mechanical adhesion but also for cell morphogenesis and differentiation. While organization of homotypic AJs is attributed mostly to classic cadherins, the adhesive mechanism of heterotypic AJs in more complex tissues remains to be clarified. Nectin, a member of a family of immunoglobulin-like adhesion molecules at various ...
متن کاملActivity-dependent alpha-cleavage of nectin-1 is mediated by a disintegrin and metalloprotease 10 (ADAM10).
Nectin-1 is known to undergo ectodomain shedding by alpha-secretase and subsequent proteolytic processing by gamma-secretase. How secretase-mediated cleavage of nectin-1 is regulated in neuronal cells and how nectin-1 cleavage affects synaptic adhesion is poorly understood. We have investigated alpha-and gamma-secretase-mediated processing of nectin-1 in primary cortical neurons and identified ...
متن کاملRole of nectin in formation of E-cadherin-based adherens junctions in keratinocytes: analysis with the N-cadherin dominant negative mutant.
E-cadherin is a Ca(2+)-dependent cell-cell adhesion molecule at adherens junctions (AJs) of epithelial cells. A fragment of N-cadherin lacking its extracellular region serves as a dominant negative mutant (DN) and inhibits cell-cell adhesion activity of E-cadherin, but its mode of action remains to be elucidated. Nectin is a Ca(2+)-independent immunoglobulin-like cell-cell adhesion molecule at ...
